ManualParticleSelection
- For each of the selected micrographs in the previous protocol, manually select the individual particles. This option can be carried out either using tilt pairs or untilted micrographs.
For untilted micrographs: Particle selection must be done in the zoom window with the left-mouse button. Zoomed area can be selected in the main windows again by clicking with the left button. Save the identified particle coordinates by typing CTRL-S or by clicking "Save Coordinates&" in the "File Menu"
For tilted pairs: Pair selection must be done in the tilted and untilted zoom windows. Save the particle coordinates in both windows (see above), and save the calculated transformation between the two micrographs by clicking "Save angles" in the "File Menu" of the untilted micrograph. Note that once the correct transformation has been found, the program accurately predicts the position of the tilted particle after identifying the untilted one.
- Input
- downsampled micrograph pair in XmippRaw format plus auxiliary file/s created in the previous protocol.
- For tilted pairs a special selection file indicating the corresponding micrograph pairs is needed. In this example with only one pair the selection file is all_pairs.sel.
In the file particle_pick.tar.gz contains all the files need for this protocol, including the all_pairs.sel and the xmipp_protocol_particle_pick.py script.
- Output
- ASCII files with particle coordinates and Euler angles assigned to each micrographs. In this example the coordinate files are down3_G40P_untilted.raw.Common.pos and down3_G40P_tilted.raw.Common.pos
File output_particle_pick.tar.gz contains the files and directory structure generated by theManual Particle Selection
protocol. To examine the contents of this file, ungzip and untar it.
