Have you tried parsing the BASE_FEE as such:
const BASE_FEE = ethers.utils.parseEther("0.25")

also it seems you have forgotten to import .env file into uploadToPinata.js as such:

const pinataSDK = require("@pinata/sdk")
const path = require("path")
const fs = require("fs")
require("dotenv").config()

If you don't import the .env file you won't be able to call API_KEY & API_SECRET from your ```uploadToPinata,js`` file
